内容記述 | Increasing numbers of nuclei and the nuclear location in the free nuclear proembryo of Ginkgo biloba were investigated. The microphotographs serial longitudinal sections of the proembryo are projected on the steel white board by a opaque projecter with halogen lamps. On the image of each nucleus, a small magnet button is placed. Photograph of the adjacent section is projected just on the former one so as to superimpose an outline of the proembryo and nuclei on those of the former. If new nuclei are projected, other magnet buttons are placed on them. And if nuclei in th former photograph disappeared, magnet buttons are removed. The numbers of magnet buttons used means preciesly the numbers of nuclei in the proembryo. The microphotographs of the serial sections of the proembryo are copied on the transparent sheet by Xerox to be enlarged 200 times. Three axes X, Y, Z are settled almost in the center of the proembryo. Location of each nucleus is measured. Then three-dimensional position of each nucleus is plotted on the set of graphs. The front view expressed by X and Y axes, the side view by Z and Y axes and the polar view by X and Z axes. After fertilization, the first nuclear division takes place obliquely to the micropyle axis in the center of the egg cell (Plate 1-A[1]). Four nuclei produced by the second division are located two-dimensionally in a plane to form a rhombus (Plate 1-D[1], D[2], D[3], D[4]; Fig.1). By the next division each of four nuclei divides to go away from the former plane. So the eight nuclei produced are lacated three-dimensionally in the proembryo (Plate 1-E[1], E[2], Fig.2; Plate 2-B[1], B[2], Fig.3). After the fourth nuclear division, sixteen nuclei spread more widely int the proembryo and they are located not homologously but somewhat to make a group (Plate 2-A[1], A[2], Fig.4; Plate 2-C, Fig.5). After the fifth and the sixth nuclear divisions, numbers of nuclei in the proembryo become slightly varied fom the value of 2[n]. After the seventh nuclear division, numbers of nuclei are also varied from 128, namely2[7]. There are some variations in the shape and the structure of nuclei and cytoplasm. Some proembryo have normal round nuclei in the granulated cytoplasm (Plate 2-G), while others have angular nuclei in the bubble-like cytoplasm (Plate 3-A[1], A[2], B[1], B[2]). The eighth nuclear division is accoumpanied by the cell wall formation. In the telophase of nuclear division an evident phragmoplast is observed at the center of the spindle (Plate 3-C[1], C[2]). Wall formations are simultaneously throughout the entire proembryo. The size of the newly formed cells is almost the same (Plate 3-D[1], D[2], Plate 4-A). Cells of the cellular proembryo do not divide synchronously (Plate 4-B[1], B[2], B[3], C[1], C[2]). |
